| STRUCTURAL AND MECHANISTIC STUDIES OF ENZYMES IN DNA REPLICATION AND REPAIR |

Our research is focused on the enzymes that are involved in the replication and repair of DNA. We utilise a variety of different techniques in the areas of molecular biology, enzymology and X-ray crystallography. We have a modern biochemistry lab equipped for a variety of techniques from gene cloning to enzymology, our own X-ray detector (Mar345 image plate) with rotating anode X-ray source (Osmic mirrors), and a variety of computer graphics workstations. The group comprises post-docs, students and technicians with a variety of expertise from molecular biology to crystallography. About half are involved in the biochemical "wet" work. This includes the molecular biology back-up for the crystallography such as cloning, overexpression and crystallisation of target proteins as well as seeking to identify functional domains of the proteins that might be more suitable for structural studies. We also undertake enzymological studies to validate mechanisms proposed from our structural studies, using techniques such as site-specific mutagenesis. The other half of the group are mainly concerned with crystallographic "dry" work. This work will often involve attempting to crystallise intermediates in the catalytic cycle of our enzymes as well as determining the initial structures of new targets. Projects will usually involve collaboration between members of the group with different areas of expertise.
